The unienzymatic nature of yeast polygalacturonase.

چکیده
Yeast polygalacturonase (YPG) catalyzes the hydrolysis of pectic acid to a mixture of digalacturonic and galacturonic acids (1). The reaction course consists of (a) an initial rapid attack on the linear chain by a random mechanism until about 25 per cent hydrolysis has occurred, (b) a slower phase during which an additional 25 per cent of the available bonds are broken, and (c) an extremely slow hydrolysis which ceases at the 70 per cent value. The reactions are as follows:

On the mechanism of action of yeast endo-polygalacturonase on oligogalacturonides and their reduced and oxidized derivatives.
Yeast polygalacturonase catalyzes a random hydrolysis of pectic acid to a mixture of digalacturonic and galacturonic acids via a series of oligogalacturonides (1). This enzyme will be termed yeast endo-polygalacturonase to differentiate it from end group-hydrolyzing enzymes, which have been named exopolygalacturonases (2). Evidence has been presented that yeast endo-polygalacturonase is a singl...
متن کاملHydrolysis of the oligogalacturonides and pectic acid by yeast polygalacturonase.
Yeast polygalacturonase (YPG) is a constitutive type enzyme, produced exocellularly and free of pectinesterase by Saccharomyces jragilis (1). It has an optimal pH of 4.4 and appears to be specific in its activity towards polygalacturonides (2). YPG differs from preparations of commercial fungus polygalacturonase in that it cannot carry out a complete breakdown of pectic acid to galacturonic aci...
متن کاملYeast Isolates From Diverse Sources For Cold- Active Polygalacturonase And Amylase Production
Cold–active polygalacturonase and amylase producers were screened using enrichment culture technique. The diverse sources screened were cold stored spoilt fruits and vegetables from different local super markets, market waste dumped soils, fruit waste dumped soils, mountain soils and Himalayan soils. About sixty yeasts showing pectinolytic activity were isolated by ruthenium red plate assay. Ei...
متن کاملPectinase and Polygalacturonase Production by a Thermophilic Aspergillus Fumigatus Isolated from Decomposting Orange Peels
A thermophilic fungal strain producing both pectinase and polygalacturonase was isolated after primary screening of 120 different isolates. The fungus was identified as Aspergillus fumigatus Fres. MTCC 4163. Using solid-state cultivation, the optimum levels of variables for pectinase and polygalacturonase (PG) production were determined. Maximal levels of enzyme activities were achieved upon gr...
متن کاملIsolation and Characterization of Polygalacturonase Produced by Tetracoccosporium sp.
Thirty-five fungal strains which isolated from vegetable wastes, were screened for the use of polygalacturonic acid as the sole carbon source. Twenty-five isolates were positive for polygalacturonase activity in cup-plate assay, as evidenced by clear hydrolysation zones. The most productive strain was determined by measuring clear zones formed around colonies stained with ruthenium red.</em...
